Inper Sight Miniaturized Microscopic Imaging System
Record cellular fluorescence in awake, freely moving animals and use companion tools for image processing, cell extraction and comparison across recordings.
Discuss configuration A small microscope. A cellular view. Inper Sight Miniaturized Microscopic Imaging System Record fluorescence in awake, freely moving animals and connect head-mounted acquisition with cell footprints and time courses. The original demonstration brings together the microscope, tether, acquisition host and software. A suitable rotary joint can manage the cable in the experimental environment. The animal moves. Imaging continues. 02 · The recording setup The compact headpiece connects to the host and comes in single-color, dual-color and stimulation-compatible configurations. An external light source and lightweight optics support freely moving imaging; dimensions and mass depend on the model. Optics in a miniature headpiece. 03 · Head-mounted optics The integrated base and GRIN lens provide optical access to the target region, supported by compatible holders and protective accessories. Select the lens for the region, preparation and microscope connection. Plan the base and lens together. 04 · Base and lens Electronic focusing spans 400 μm for adjusting the observed plane after assembly. Multi-plane acquisition requires a compatible configuration; focus range is not the accessible depth of a brain region. Focus from the workspace. 05 · Electronic focusing Spectral and stimulation modes depend on the model; check crosstalk and timing in dual-channel experiments. Sight 110 provides green imaging; Sight 120 adds an optical stimulation option. Green Green fluorescence Sight 210 provides red imaging; Sight 220 combines red imaging with optical stimulation. Red Red fluorescence Sight 310 provides red/green imaging; Sight 320 adds optical stimulation. Select according to the probes and spectra. Dual color Compare two fluorescence channels Match the configuration to the fluorophore. The acquisition workspace displays the fluorescence image and intensity histogram to help check exposure, brightness distribution and sampling. Inspect a local field in more detail rather than judging quality from an average trace alone. Inspect the view before recording. 07 · Field-of-view inspection Draw rectangular, circular or polygonal ROIs and follow fluorescence or ΔF/F in real time. Live ROI traces aid acquisition monitoring; offline cell extraction uses the analysis workflow. Draw a region. Follow its signal. 08 · Live ROIs Companion behavior video and event markers preserve experimental context. Configure internal/external triggers and DIO, using manual or signal-conditioned markers, then verify timing before the experiment. Bring behavior and stimuli onto the imaging timeline. 09 · Synchronized events These images come from the original materials. Field, cell count and depth depend on lens, expression and preparation and are not fixed outcomes for every sample. Inspect fluorescent structures and background in the source field before choosing analysis parameters. Hippocampus HPC example Use a GRIN lens suited to the target location for deeper-region imaging, with the preparation confirmed for the experiment. Hypothalamus VMH example Inspect real views from different regions. Companion analysis moves through preprocessing, candidate-cell initialization and spatial/temporal updates. Results link footprints to time courses for cell-by-cell review. From a video to cells and traces. 11 · Analysis overview Use deglow, denoising, background removal, motion correction and cropping as appropriate, checking processing strength with Test. Choose a Median or Mean window with cell scale in mind and inspect for oversmoothed boundaries. Denoise Reduce noise while inspecting structure Choose the window for background and cell scale, checking that weak targets are not removed. DeBg Reduce slowly varying background Inspect the field and residual movement before and after correction, cropping the valid region when needed. Motion correction Align frame-to-frame displacement Prepare the images before finding cells. Candidate filtering connects image preparation and initialization. Check thresholds against the temporal signal. Generate candidate cell centers for quality filtering and footprint reconstruction. Candidate count is not the final cell count. Candidate centers Find candidates with DotFind Use peak-to-noise ratio, statistical filtering and Test traces to review candidates, balancing weak-signal loss against noise retention. Quality filtering Review candidates with PNR and Ks Find candidates. Then assess quality. DotMerge organizes candidates using distance and temporal correlation; Rebuilt forms initial spatial and temporal matrices, and C-Merge checks duplicate units. Review traces and background to avoid merging neighboring cells incorrectly. Build initial footprints from the centers. 14 · Initialization Noise estimation, spatial updates, temporal updates and final merging lead to CNMF results. Parameters affect retained footprints and traces; inspect them against source data. Inferred events are not directly measured action potentials. Refine footprints and time courses together. 15 · Spatial and temporal updates CrossReg matches different CNMF results to help inspect cell correspondence across trials or days. Input dimensions must agree; review distance thresholds and conflicting matches. Compare recordings with spatial correspondence. 16 · Cross-registration Use ΔF/F for relative fluorescence change or Z-Score for standardized deviation. Group trials with markers and event windows and document the baseline alongside traces and heatmaps. Compare activity against a baseline. 17 · Normalization and grouping Save routine settings and recheck spectra, planes and triggers before repeating a run. Acquisition settings Export processed data and results from the relevant modules for subsequent statistics and figures. Analysis export Choose the system around region, probe, movement environment and analysis needs. Configuration matching Keep the process and reviewable results.
